Description: | SHP2 inhibitor LY6 (LY6) is a small molecule inhibitor that stabilizes the autoinhibited conformation of the oncogenic tyrosine phosphatase SHP2, inhibits SHP2 with IC50 of 9.8 uM, 7-fold selectivity over SHP1; inhibits full length WT SHP2, full length SHP2 E76K, and the SHP2 PTP domain with IC50 of 9.8, 7.67 and20.87 uM, respectively; effectively inhibits SHP2-mediated cell signaling and proliferation, mouse and patient leukemia cells with PTPN11 activating mutations were more sensitive to this inhibitor than wild-type cells. |
