Description: | SynuClean-D (SC-D) is a novel, small molecule inhibitor of α-synuclein (α-Syn) aggregation, incubation of 70 uM α-Syn with 100 uM SC-D impacted α-Syn aggregation in vitro, as monitored by Th-T fluorescence; significantly reduces the in vitro aggregation of wild-type α-synuclein and the familiar A30P and H50Q variants in a substoichiometric molar ratio, prevents fibril propagation in protein-misfolding cyclic amplification assays and decreases the number of α-synuclein inclusions in human neuroglioma cells; binds to cavities in mature α-synuclein fibrils and displays a strong fibril disaggregation activity; significantly reduces the toxicity exerted by α-synuclein, also rescues dopaminergic neurons from α-synuclein-induced degeneration in Parkinson's disease models. |
